Les personnes en bonne sant\u00e9 qui nous entourent respirent ces gouttelettes contenant l\u2019agent pathog\u00e8ne, qui se fixent \u00e0 l\u2019int\u00e9rieur de leur nez et infectent les cellules qui tapissent les voies nasales.<\/p>\n<p>L\u2019agent pathog\u00e8ne se r\u00e9plique et peut \u00eatre lib\u00e9r\u00e9 dans l\u2019air lorsqu\u2019une personne malade, qu\u2019elle le sache ou non, \u00e9ternue, tousse, rit, chante ou m\u00eame respire simplement.<\/p>\n<p>La nouvelle \u00e9tude d\u00e9taille les efforts de l\u2019\u00e9quipe de recherche pour cr\u00e9er un spray nasal pour se d\u00e9fendre contre les maladies respiratoires a\u00e9roport\u00e9es.<\/p>\n<p>\u00ab Le spray, appel\u00e9 Pathogen Capture and Neutralizing Spray (PCANS) dans l&rsquo;article, a \u00e9t\u00e9 d\u00e9velopp\u00e9 \u00e0 l&rsquo;aide d&rsquo;ingr\u00e9dients de la base de donn\u00e9es des ingr\u00e9dients inactifs (IID) de la FDA, qui ont d\u00e9j\u00e0 \u00e9t\u00e9 utilis\u00e9s dans des sprays nasaux approuv\u00e9s, ou de la liste g\u00e9n\u00e9ralement reconnue comme s\u00fbre (GRAS) de la FDA \u00bb, a d\u00e9clar\u00e9 le co-auteur principal Nitin Joshi, Ph.D., professeur adjoint d&rsquo;anesth\u00e9siologie au Brigham and Women&rsquo;s Hospital.<\/p>\n<p>\u00ab Nous avons d\u00e9velopp\u00e9 une formulation sans m\u00e9dicament utilisant ces compos\u00e9s pour bloquer les germes de trois mani\u00e8res : le PCANS forme une matrice semblable \u00e0 un gel qui pi\u00e8ge les gouttelettes respiratoires, immobilise les germes et les neutralise efficacement, pr\u00e9venant ainsi l&rsquo;infection. \u00bb<\/p>\n<p>Les chercheurs ont r\u00e9alis\u00e9 les exp\u00e9riences d\u00e9taill\u00e9es dans l\u2019\u00e9tude en laboratoire. Ils n\u2019ont pas \u00e9tudi\u00e9 le PCANS directement chez l\u2019homme. Les chercheurs ont d\u00e9velopp\u00e9 la formulation et \u00e9tudi\u00e9 sa capacit\u00e9 \u00e0 capturer les gouttelettes respiratoires dans une r\u00e9plique imprim\u00e9e en 3D d\u2019un nez humain. Ils ont montr\u00e9 que lorsqu\u2019il \u00e9tait pulv\u00e9ris\u00e9 dans la r\u00e9plique de la cavit\u00e9 nasale, le PCANS capturait deux fois plus de gouttelettes que le mucus seul.<\/p>\n<p>\u00ab Le PCANS forme un gel, augmentant sa r\u00e9sistance m\u00e9canique d&rsquo;un facteur cent, formant une barri\u00e8re solide \u00bb, a d\u00e9clar\u00e9 l&rsquo;auteur principal John Joseph, Ph.D., ancien chercheur postdoctoral au Brigham and Women&rsquo;s Hospital. \u00ab Il a bloqu\u00e9 et neutralis\u00e9 presque 100 % de tous les virus et bact\u00e9ries que nous avons test\u00e9s, y compris la grippe, le SARS-CoV-2, le VRS, l&rsquo;ad\u00e9novirus, la pneumonie K et bien d&rsquo;autres. \u00bb<\/p>\n<p>Des exp\u00e9riences sur des souris ont montr\u00e9 qu&rsquo;une dose unique de vaporisateur nasal PCANS pouvait bloquer efficacement l&rsquo;infection par un virus de la grippe (PR8) \u00e0 une dose 25 fois sup\u00e9rieure \u00e0 la dose mortelle. Les niveaux de virus dans les poumons ont \u00e9t\u00e9 r\u00e9duits de plus de 99,99 %, et les cellules inflammatoires et les cytokines dans les poumons des animaux trait\u00e9s par PCANS \u00e9taient normales.<\/p>\n<p>\u00ab La capacit\u00e9 de la formulation \u00e0 inactiver un large spectre d&rsquo;agents pathog\u00e8nes, y compris le virus mortel de la grippe PR8, d\u00e9montre sa grande efficacit\u00e9 \u00bb, a d\u00e9clar\u00e9 le co-auteur principal Yohannes Tesfaigzi, Ph.D., professeur de m\u00e9decine AstraZeneca dans le domaine des maladies respiratoires et inflammatoires au Brigham and Women&rsquo;s Hospital.<\/p>\n<p>\u00ab Dans une \u00e9tude rigoureuse sur un mod\u00e8le murin, le traitement prophylactique avec PCANS a d\u00e9montr\u00e9 une efficacit\u00e9 exceptionnelle, les souris trait\u00e9es pr\u00e9sentant une protection compl\u00e8te, tandis que le groupe non trait\u00e9 n&rsquo;a montr\u00e9 aucun avantage de ce type. \u00bb<\/p>\n<p>Bien que l&rsquo;\u00e9tude soit limit\u00e9e par l&rsquo;absence d&rsquo;\u00e9tudes humaines sur le PCANS, elle fournit une base solide pour de futures recherches visant \u00e0 explorer le plein potentiel du PCANS dans un contexte plus large.
