Les anticorps uniques ciblent un site essentiel hautement conserv\u00e9 \u00e0 la base de la prot\u00e9ine de pointe du virus, le serrant efficacement et emp\u00eachant le virus d&rsquo;infecter les cellules.<\/p>\n<p>Les r\u00e9sultats, publi\u00e9s dans <i>Communications de la nature<\/i>offrez une voie prometteuse pour d\u00e9velopper des traitements antiviraux \u00e0 large spectre qui pourraient rester efficaces contre les futures variantes virales.<\/p>\n<p>SARS-COV-2, le virus derri\u00e8re Covid-19, continue d&rsquo;\u00eatre une menace potentielle \u00e0 mesure qu&rsquo;elle \u00e9volue vers de nouvelles variantes r\u00e9sistantes aux th\u00e9rapies anticorps actuellement approuv\u00e9es. La r\u00e9sistance \u00e9merge largement parce que les anticorps ciblent g\u00e9n\u00e9ralement les r\u00e9gions virales, telles que le domaine de liaison aux r\u00e9cepteurs de la prot\u00e9ine de pointe, qui mute \u00e9galement fr\u00e9quemment, permettant d&rsquo;\u00e9chapper \u00e0 la reconnaissance des anticorps.<\/p>\n<p>Pour y rem\u00e9dier, une \u00e9quipe de recherche dirig\u00e9e par le professeur Xavier Saelens et le Dr Bert Schepens au VIB-Ugent Center for Medical Biotechnology ont explor\u00e9 une strat\u00e9gie diff\u00e9rente en se concentrant sur l&rsquo;une des sous-unit\u00e9s les plus stables de la prot\u00e9ine Spike. La soi-disant sous-unit\u00e9 S2 est essentielle pour la capacit\u00e9 du virus \u00e0 fusionner avec les cellules h\u00f4tes, un processus essentiel pour l&rsquo;infection, et il est plus conserv\u00e9 dans diff\u00e9rents coronavirus.<\/p>\n<h2>Une pince mol\u00e9culaire sur le virus<\/h2>\n<p>L&rsquo;\u00e9quipe s&rsquo;est tourn\u00e9e vers les lamas (plus sp\u00e9cifiquement un lama appel\u00e9 Winter). Les lamas g\u00e9n\u00e8rent des anticorps dits de domaine unique, \u00e9galement connus sous le nom de VHHS ou nanobodies, qui sont beaucoup plus petits que les anticorps g\u00e9n\u00e9r\u00e9s par la plupart des animaux, y compris les humains. Les chercheurs ont identifi\u00e9 plusieurs anticorps LLAMA qui neutralisent fortement un large panel de coronavirus du SRAS.<\/p>\n<p>Ce qui rend ces anticorps particuli\u00e8rement prometteurs, c&rsquo;est leur mode d&rsquo;action unique: ils agissent comme une pince mol\u00e9culaire. Ils s&rsquo;accrochent \u00e0 la r\u00e9gion mal expos\u00e9e et hautement conserv\u00e9e (une bobine enroul\u00e9e de trois h\u00e9lices alpha) \u00e0 la base de la prot\u00e9ine de pointe du virus. Ce faisant, ils verrouillent la prot\u00e9ine de pointe dans sa forme d&rsquo;origine, l&#8217;emp\u00eachant physiquement de se d\u00e9rouler sous la forme dont le virus a besoin pour infecter les cellules.<\/p>\n<p>Les anticorps ont montr\u00e9 une forte protection contre l&rsquo;infection chez les animaux de laboratoire, m\u00eame \u00e0 faible doses. Et lorsque les chercheurs ont tent\u00e9 de forcer le virus \u00e0 \u00e9voluer la r\u00e9sistance, le virus a lutt\u00e9, ne produisant que des variantes d&rsquo;\u00e9vasion rares qui \u00e9taient beaucoup moins contagantes.
