Les r\u00e9sultats de l&rsquo;\u00e9tude peuvent d\u00e9bloquer de nouvelles voies pour comprendre et traiter les troubles intestinaux complexes.<\/p>\n<p>La prot\u00e9ine agit comme un capteur interne dans les cellules musculaires lisses de l&rsquo;intestin. Il aide les intestins \u00e0 savoir quand et comment d\u00e9placer les aliments dans le syst\u00e8me digestif.<\/p>\n<p>L&rsquo;intestin est m\u00e9caniquement tr\u00e8s actif. Son tube digestif se contracte et se d\u00e9tend constamment. La contraction rythmique des cellules musculaires lisses aide \u00e0 d\u00e9composer les aliments et \u00e0 absorber les nutriments. Les perturbations de ce processus conduisent \u00e0 une gamme de troubles de la motilit\u00e9 intestinale.<\/p>\n<p>Mais comment les cellules musculaires de l&rsquo;intestin savent-elles quand se contracter?<\/p>\n<p>C&rsquo;est la question que les chercheurs de l&rsquo;UC Davis et de l&rsquo;UCLA ont d\u00e9cid\u00e9 de r\u00e9pondre \u00e0 travers une s\u00e9rie de tests dans des mod\u00e8les de souris. Ils ont concentr\u00e9 leur \u00e9tude sur PIEZO1. Leur travail a \u00e9t\u00e9 publi\u00e9 dans <i>Biologie des communications<\/i>.<\/p>\n<h2>D\u00e9bullation, signalisation et r\u00e9gulation des cellules musculaires dans l&rsquo;intestin<\/h2>\n<p>Piezo1 est connu pour d\u00e9tecter les forces physiques, telles que l&rsquo;\u00e9tirement et la pression. Lorsque les chercheurs ont g\u00e9n\u00e9tiquement retir\u00e9 le pi\u00e9zo1 des cellules musculaires lisses intestinales, les effets \u00e9taient dramatiques: perte de poids, transit alimentaire retard\u00e9 et amincissement des couches musculaires intestinales. Ils ont \u00e9galement d\u00e9tect\u00e9 une perturbation significative de la signalisation du calcium essentielle pour une bonne contraction musculaire.<\/p>\n<p>\u00ab\u00a0Les r\u00e9sultats ont \u00e9t\u00e9 frappants. Malgr\u00e9 les changements profonds de leur motilit\u00e9 intestinale, ces souris ont continu\u00e9 \u00e0 vivre aussi longtemps que d&rsquo;autres souris normales. Cette pr\u00e9sence semble aider \u00e0 contr\u00f4ler le passage des ions calcium &#8211; les joueurs cl\u00e9s en cas de muscle\u00a0\u00bb, a expliqu\u00e9 l&rsquo;auteur senior de l&rsquo;\u00e9tude, Mart\u00edn G. Mart\u00edn. Il est professeur de gastro-ent\u00e9rologie p\u00e9diatrique \u00e0 l&rsquo;UCLA.<\/p>\n<p>Le positionnement interne de Piezo1 sugg\u00e8re qu&rsquo;il peut \u00eatre impliqu\u00e9 dans les m\u00e9canismes de signalisation du calcium critiques pour le rythme et la force appropri\u00e9s g\u00e9n\u00e9r\u00e9s lors des contractions intestinales. Piezo1 peut \u00eatre un r\u00e9gulateur auparavant m\u00e9connu de la sant\u00e9 intestinale\u00a0\u00bb, a d\u00e9clar\u00e9 Bautista.<\/p>\n<p>Bautista \u00e9tudie actuellement le r\u00f4le de Piezo1 pendant le d\u00e9veloppement de l&rsquo;intestin f\u0153tal. Son \u00e9quipe \u00e9tudie comment son absence affecte la motilit\u00e9 et la fonction de l&rsquo;intestin apr\u00e8s la naissance. Leurs recherches peuvent offrir des informations sur de nombreuses conditions intestinales postnatales, comme la gastroschisie, l&rsquo;ent\u00e9rocolite n\u00e9crosante (NEC) et le syndrome de l&rsquo;intestin court.
