L&rsquo;agent pathog\u00e8ne qui provoque la maladie, Mycobacterium tuberculosis, est mortel en partie en raison de son enveloppe externe complexe, ce qui l&rsquo;aide \u00e0 \u00e9chapper aux r\u00e9ponses immunitaires des h\u00f4tes infect\u00e9s.<\/p>\n<p>Dans un <i>ACS Maladies infectieuses<\/i> Document, les chercheurs ont d\u00e9velopp\u00e9 une sonde chimique pour \u00e9tudier une composante cl\u00e9 de cette enveloppe. Leurs r\u00e9sultats fournissent une \u00e9tape vers la recherche de nouvelles fa\u00e7ons d&rsquo;inactiver la bact\u00e9rie.<\/p>\n<p>Parce que la gu\u00e9rison de la tuberculose n\u00e9cessite de prendre des m\u00e9dicaments pendant des mois, ce qui peut entra\u00eener une r\u00e9sistance \u00e0 la tuberculose \u00e0 certains antibiotiques, les scientifiques travaillent \u00e0 d\u00e9velopper de nouveaux traitements. Une cible possible est la couche la plus externe de la bact\u00e9rie, appel\u00e9e Mycomembrane, qui prot\u00e8ge les bact\u00e9ries des facteurs de stress. Lorsque M. tuberculosis est attaqu\u00e9 par les cellules immunitaires des macrophages d&rsquo;un h\u00f4te, le mycomembrane produit des compos\u00e9s qui suppriment la r\u00e9ponse immunitaire de l&rsquo;h\u00f4te infect\u00e9.<\/p>\n<p>Auparavant, Ben Swarts, Sloan Siegrist et ses coll\u00e8gues ont d\u00e9velopp\u00e9 des sondes chimiques activ\u00e9es par la lumi\u00e8re qui imitent certains de ces compos\u00e9s et ont \u00e9tudi\u00e9 comment ils pourraient interagir avec les cellules immunitaires d&rsquo;un h\u00f4te. Maintenant, les chercheurs ont d\u00e9velopp\u00e9 une sonde chimique pour une autre composante Mycomembrane appel\u00e9e acide mycolique. Ils ont con\u00e7u la sonde d&rsquo;acide mycolique pour permettre la capture de prot\u00e9ines interagissantes dans les cellules immunitaires de l&rsquo;h\u00f4te &#8211; des informations qui pourraient aider les scientifiques \u00e0 mieux comprendre comment M. tuberculosis survit dans l&rsquo;environnement d&rsquo;assaut de l&rsquo;h\u00f4te.<\/p>\n<p>Dans un immunodosage enzymatique, la sonde a stimul\u00e9 une r\u00e9ponse immunitaire similaire \u00e0 l&rsquo;acide mycolique r\u00e9el dans les cellules de macrophages cultiv\u00e9es de la souris. En utilisant le balayage de fluorescence, les chercheurs ont observ\u00e9 que les prot\u00e9ines photo-marqu\u00e9es \u00e0 sonde \u00e0 sonde mycolique dans les cellules de macrophages.
