DOI: 10.1038\/s42003-025-07973-5\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>Une nouvelle application pour un compos\u00e9 appel\u00e9 ML233 a montr\u00e9 des r\u00e9sultats prometteurs pour inhiber la production de m\u00e9lanine, offrant de nouvelles strat\u00e9gies potentielles pour le traitement des affections cutan\u00e9es li\u00e9es aux pigments et au moins un type de m\u00e9lanome.<\/p>\n<p>Publi\u00e9 dans la revue <i>Biologie des communications<\/i>la recherche marque une \u00e9tape cruciale vers la lutte contre un besoin non satisfait en dermatologie. Il a \u00e9t\u00e9 men\u00e9 sous les auspices de MDI Bioscience, une initiative de d\u00e9couverte de m\u00e9dicaments du MDI Biological Laboratory.<\/p>\n<p>Dans de nombreuses esp\u00e8ces, la m\u00e9lanine est responsable de la coloration de la peau, des cheveux et de l&rsquo;iris. La m\u00e9lanine est synth\u00e9tis\u00e9e dans des cellules sp\u00e9cialis\u00e9es appel\u00e9es m\u00e9lanocytes, dans un processus appel\u00e9 m\u00e9lanogen\u00e8se. Les d\u00e9fauts de la m\u00e9lanogen\u00e8se sont associ\u00e9s \u00e0 des troubles cutan\u00e9s tels que l&rsquo;albinisme, les troubles de l&rsquo;hyperpigmentation tels que le vitiligo et le m\u00e9lasma et le m\u00e9lanome.<\/p>\n<p>Les troubles de l&rsquo;hyperpigmentation peuvent avoir un impact sur la qualit\u00e9 de vie des patients en raison de l&rsquo;apparence alt\u00e9r\u00e9e, et leur traitement repr\u00e9sente un march\u00e9 mondial pr\u00e9vu pour atteindre environ 11,84 milliards de dollars d&rsquo;ici 2033. Malgr\u00e9 des d\u00e9cennies de recherche, aucun traitement efficace n&rsquo;existe actuellement qui affecte la m\u00e9lanogen\u00e8se sans effets secondaires ind\u00e9sirables.<\/p>\n<p>Dirig\u00e9e par l&rsquo;investigateur de laboratoire biologique MDI, Romain Madelaine, Ph.D., la nouvelle \u00e9tude a caract\u00e9ris\u00e9 le ML233 comme un inhibiteur puissant et direct de la tyrosinase, une enzyme qui r\u00e9gule le rythme de la m\u00e9lanogen\u00e8se. Test\u00e9 dans un mod\u00e8le de poisson z\u00e8bre vivant et dans les cellules cultiv\u00e9es en laboratoire de souris et d&rsquo;humains, le compos\u00e9 a effectivement r\u00e9duit la production de m\u00e9lanine sans effets secondaires toxiques significatifs observ\u00e9s.<\/p>\n<p>\u00ab\u00a0Nos r\u00e9sultats sugg\u00e8rent que ML233 interagit directement avec le site actif de la tyrosinase, inhibant sa fonction et r\u00e9duisant finalement la synth\u00e8se de la m\u00e9lanine\u00a0\u00bb, a d\u00e9clar\u00e9 Madelaine. \u00ab\u00a0Ce m\u00e9canisme met en \u00e9vidence le ML233 comme une approche potentiellement efficace et bien tol\u00e9r\u00e9e pour traiter les affections cutan\u00e9es associ\u00e9es aux m\u00e9lanocytes.\u00a0\u00bb<\/p>\n<p>Les r\u00e9sultats ouvrent de nouvelles voies pour le d\u00e9veloppement de th\u00e9rapies \u00e0 base de ML233 qui pourraient am\u00e9liorer consid\u00e9rablement les r\u00e9sultats pour les personnes touch\u00e9es par les troubles de la pigmentation. D&rsquo;autres \u00e9tudes seront n\u00e9cessaires pour \u00e9valuer l&rsquo;innocuit\u00e9 \u00e0 long terme et l&rsquo;efficacit\u00e9 de ML233 en milieu clinique.<\/p>\n<p>\u00ab\u00a0La nouveaut\u00e9 de nos r\u00e9sultats n&rsquo;est pas n\u00e9cessairement dans la biologie elle-m\u00eame, mais dans la qualit\u00e9, l&rsquo;efficacit\u00e9 et les faibles effets secondaires observ\u00e9s avec ce compos\u00e9 chimique\u00a0\u00bb, a d\u00e9clar\u00e9 Madelaine. \u00ab\u00a0Nous avons observ\u00e9 de forts effets sur la m\u00e9lanogen\u00e8se \u00e0 des doses tr\u00e8s faibles.
