Les microbes associ\u00e9s \u00e0 ces infections entrent dans un stade latent au cours duquel ils se cachent tranquillement dans les cellules, jouant au long jeu pour \u00e9chapper \u00e0 la capture et assurer leur propre survie. Mais un manque de mod\u00e8les naturels pour \u00e9tudier ces \u00e9tapes au repos a conduit \u00e0 des lacunes dans la compr\u00e9hension des scientifiques de la fa\u00e7on dont la latence contribue \u00e0 la persistance des agents pathog\u00e8nes et si ces \u00e9tapes peuvent \u00eatre cibl\u00e9es par le syst\u00e8me immunitaire.<\/p>\n<p>Maintenant, une \u00e9quipe dirig\u00e9e par des chercheurs de l&rsquo;Universit\u00e9 de Pennsylvania en m\u00e9decine v\u00e9t\u00e9rinaire montre que le syst\u00e8me immunitaire reconna\u00eet en effet le stade latent du parasite Toxoplasma gondii, qui provoque une toxoplasmose &#8211; r\u00e9duisant certaines hypoth\u00e8ses communes sur la fa\u00e7on dont le syst\u00e8me immunitaire traite des infections dans le cerveau.<\/p>\n<p>L&rsquo;\u00e9tude est publi\u00e9e dans <i>Microbiologie de la nature<\/i>.<\/p>\n<p>Le professeur de Penn Vet Christopher A. Hunter, l&rsquo;auteur principal du journal, a d\u00e9clar\u00e9 que cette connaissance soutient l&rsquo;id\u00e9e que les kystes de toxoplasma gondii peuvent \u00eatre cibl\u00e9s et peut-\u00eatre m\u00eame effac\u00e9s, et les r\u00e9sultats ont des implications pour d&rsquo;autres infections et th\u00e9rapies futures potentielles. Le document montre \u00e9galement comment les kystes favorisent la survie mutuelle du parasite et de l&rsquo;h\u00f4te.<\/p>\n<p>Dans son stade latent, Toxoplasma gondii forme des kystes \u00e0 longue dur\u00e9e de vie dans les neurones du cerveau, ce qui aide le parasite \u00e0 \u00e9chapper \u00e0 la r\u00e9ponse immunitaire de l&rsquo;h\u00f4te. Cependant, dans cette \u00e9tude, les chercheurs ont constat\u00e9 que certaines cellules T peuvent cibler les neurones contenant des kystes, favorisant ainsi le contr\u00f4le des parasites. Mais il y a un compromis: ils ont \u00e9galement constat\u00e9 que lorsque les kystes ne sont pas form\u00e9s, il y a une charge de parasite encore plus \u00e9lev\u00e9e et des dommages accrus au cerveau.<\/p>\n<p>\u00ab\u00a0Il y a cet \u00e9quilibre entre les agents pathog\u00e8nes qui doivent s&rsquo;installer dans l&rsquo;h\u00f4te mais ne pas d\u00e9velopper autant que cela est pr\u00e9judiciable \u00e0 l&rsquo;h\u00f4te, car si l&rsquo;h\u00f4te d\u00e9c\u00e8de, l&rsquo;agent pathog\u00e8ne peut ne pas survivre\u00a0\u00bb, explique l&rsquo;auteur Lindsey A. BlAlberg, qui, au moment de la recherche, \u00e9tait un doctorant du laboratoire de Hunter.<\/p>\n<p>Le toxoplasma gondii provoque une toxoplasmose, une infection asymptomatique pour la plupart des personnes en bonne sant\u00e9 mais pr\u00e9sente un plus grand risque pour ceux qui sont immunod\u00e9prim\u00e9s ou enceintes. Elle est caus\u00e9e par la consommation de viande contamin\u00e9e et mal cuite et par l&rsquo;exposition \u00e0 des excr\u00e9ments de chats infect\u00e9s, car les f\u00e9lins sont le seul animal dans lequel le parasite peut se reproduire sexuellement.<\/p>\n<p>La co-auteure Julia N. Eberhard, \u00e9tudiante doctorale en immunologie, souligne deux r\u00e9sultats qui vont \u00e0 l&rsquo;encontre de la litt\u00e9rature pr\u00e9existante et des notions communes parmi les immunologues. Elle dit que les scientifiques ont longtemps pens\u00e9 que les kystes de toxoplasma gondii pouvaient se cacher dans les neurones pour pr\u00e9venir la reconnaissance immunitaire, mais cette \u00e9tude a montr\u00e9 que \u00ab\u00a0les neurones ne sont pas un refuge complet pour les agents pathog\u00e8nes\u00a0\u00bb.<\/p>\n<p>Eberhard dit qu&rsquo;une autre croyance commun\u00e9ment d\u00e9tenue \u00e9tait que le parasite doit former des kystes pour pouvoir persister, mais en regardant une souche de parasite qui ne pouvait pas se convertir au stade du kyste, les chercheurs ont constat\u00e9 que le syst\u00e8me immunitaire ne d\u00e9gageait pas le parasite. Ils pouvaient encore identifier les parasites chez la souris six mois plus tard, ce qu&rsquo;Eberhard a trouv\u00e9 tr\u00e8s surprenant.<\/p>\n<p>La mod\u00e9lisation math\u00e9matique a confirm\u00e9 ind\u00e9pendamment les r\u00e9sultats exp\u00e9rimentaux et a indiqu\u00e9 que la pression immunitaire sur le stade latent de Toxoplasma gondii pourrait expliquer la mont\u00e9e et la chute observ\u00e9es en nombre de kystes.
