Hurd, Ph.D., D\u00e9partements de neurosciences et de psychiatrie, Friedman Brain Institute, ICAHN School of Medicine du Mount Sinai et Addiction Institute of Mount Sina\u00ef, New York, \u00ab\u00a0Mon \u00e9quipe de recherche et moi sommes amen\u00e9s \u00e0 \u00e9tendre les perspicaces neurobiologiques sous-jacentes \u00e0 la d\u00e9pendance.<\/p>\n<p>\u00ab\u00a0En examinant les signatures mol\u00e9culaires dans le cerveau des utilisateurs d&rsquo;opio\u00efdes humains, nous esp\u00e9rons mieux comprendre la neuropathologie du trouble de la consommation d&rsquo;opio\u00efdes au-del\u00e0 des m\u00e9canismes de r\u00e9compense aigus et d&rsquo;identifier de nouvelles voies de traitement pertinentes pour les ph\u00e9notypes centraux qui perp\u00e9tuent la consommation de substances.\u00a0\u00bb<\/p>\n<p>Dans cette \u00e9tude, l&rsquo;\u00e9quipe de recherche a appliqu\u00e9 l&rsquo;apprentissage automatique pour distinguer la signature mol\u00e9culaire dans le cortex orbitofrontal, une r\u00e9gion du cerveau critique pour les aspects du contr\u00f4le des impulsions, le comportement de recherche de m\u00e9dicaments et les fonctions cognitives li\u00e9es aux troubles de la consommation de substances.<\/p>\n<p>Les enqu\u00eateurs ont constat\u00e9 que l&rsquo;algorithme d&rsquo;apprentissage automatique n&rsquo;\u00e9tait pas seulement efficace pour identifier les signatures distinguant le cerveau d&rsquo;un utilisateur d&rsquo;h\u00e9ro\u00efne humain, mais il a \u00e9galement identifi\u00e9 le g\u00e8ne le plus pr\u00e9dictif de cette signature mol\u00e9culaire appel\u00e9e Shisa7 qui n&rsquo;avait pas \u00e9t\u00e9 explor\u00e9e pr\u00e9c\u00e9demment dans le domaine. Une enqu\u00eate plus approfondie a r\u00e9v\u00e9l\u00e9 que la modulation de l&rsquo;expression de ce g\u00e8ne dans le cortex orbitofrontal influence le comportement de recherche d&rsquo;h\u00e9ro\u00efne et la flexibilit\u00e9 cognitive.<\/p>\n<p>Le Dr Hurd ajoute: \u00ab\u00a0Nous avons \u00e9galement observ\u00e9 que lorsque nous avons surexprim\u00e9 SHISA7 chez les animaux na\u00effs de m\u00e9dicaments, il a compl\u00e8tement issu la signature du transcriptome observ\u00e9e avec une utilisation r\u00e9p\u00e9t\u00e9e de l&rsquo;h\u00e9ro\u00efne.
