Credit: Zhu Cancan\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>Une \u00e9quipe de chercheurs des Instituts des sciences physiques de l&rsquo;Acad\u00e9mie des sciences chinoises a d\u00e9velopp\u00e9 un syst\u00e8me de tests de point de service (POCT) bas\u00e9 sur la technologie microfluidique CRISPR-DX, am\u00e9liorant consid\u00e9rablement la sensibilit\u00e9, la vitesse et la capacit\u00e9 de multiplexage du pathog\u00e8ne d\u00e9tection.<\/p>\n<p>Les r\u00e9sultats ont \u00e9t\u00e9 publi\u00e9s dans <i>Biocapteurs et bio\u00e9lectronique<\/i>.<\/p>\n<p>Les diagnostics mol\u00e9culaires \u00e0 base de CRISPR \/ CAS peuvent d\u00e9tecter avec pr\u00e9cision des s\u00e9quences de g\u00e8nes sp\u00e9cifiques dans les agents pathog\u00e8nes, notamment des mutations g\u00e9n\u00e9tiques et des anomalies, fournissant des informations importantes pour le diagnostic et le traitement de la maladie.<\/p>\n<p>Bien qu&rsquo;il offre une pr\u00e9cision \u00e9lev\u00e9e et est consid\u00e9r\u00e9e comme une alternative prometteuse \u00e0 la r\u00e9action en cha\u00eene par polym\u00e9rase, il a des limites telles que une faible sensibilit\u00e9, une capacit\u00e9 limit\u00e9e et le besoin de plusieurs instruments de laboratoire, ce qui le rend difficile pour une d\u00e9tection multi-pathog\u00e8ne rapide et pratique dans le point de vue Param\u00e8tres de soins.<\/p>\n<p>Pour relever ces d\u00e9fis, les chercheurs ont combin\u00e9 des puces microfluidiques avec la technologie CRISPR \/ CAS12A pour cr\u00e9er un syst\u00e8me \u00e0 double \u00e9tage qui s\u00e9pare les processus d&rsquo;amplification et de d\u00e9tection. Cette s\u00e9paration, ainsi qu&rsquo;une m\u00e9thode de centrifugation en deux \u00e9tapes, emp\u00eache la contamination pendant les tests.<\/p>\n<p>Le syst\u00e8me permet \u00e9galement des canaux de d\u00e9tection personnalisables en fonction du nombre d&rsquo;agents pathog\u00e8nes, am\u00e9liorant la sensibilit\u00e9 et la capacit\u00e9 de tester plusieurs agents pathog\u00e8nes \u00e0 la fois.<\/p>\n<p>Le syst\u00e8me d&rsquo;analyse POCT Lift-CM, qui comprend une plate-forme de chauffage, un module de centrifugation et une d\u00e9tection de fluorescence en temps r\u00e9el. Il est compact, facile \u00e0 utiliser et contr\u00f4l\u00e9 par une application pour smartphone, r\u00e9duisant le besoin de plusieurs instruments de laboratoire.
