Credit: Prof. Eunjung Kim from INU, Korea, https:\/\/doi.org\/10.1016\/j.snb.2024.136933\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>L\u2019\u00e9pid\u00e9mie de COVID-19 en 2019 a d\u00e9clench\u00e9 des mesures visant \u00e0 sensibiliser le public aux pand\u00e9mies et a \u00e9galement conduit \u00e0 un d\u00e9veloppement acc\u00e9l\u00e9r\u00e9 de vaccins. Si ces mesures ont contribu\u00e9 \u00e0 r\u00e9duire consid\u00e9rablement la transmission virale, elles ont \u00e9galement eu des cons\u00e9quences inattendues, telles qu\u2019une r\u00e9duction globale de la propagation d\u2019autres virus, entra\u00eenant des pauses dans les r\u00e9gimes de vaccination. Cependant, les agents pathog\u00e8nes \u00e0 mutation rapide comme les virus repr\u00e9sentent toujours une menace importante, avec des pr\u00e9dictions de co-infections avec plusieurs virus provoquant des \u00ab twind\u00e9mies \u00bb ou des \u00ab tripled\u00e9mies \u00bb dans le futur.<\/p>\n<p>Les PCR quantitatives \u00e0 la transcriptase inverse (RT-qPCR) sont des tests de diagnostic de maladies fiables, mais sont limit\u00e9es par l&rsquo;utilisation d&rsquo;\u00e9quipements et de r\u00e9actifs co\u00fbteux, ce qui limite leur utilit\u00e9 dans des contextes aux ressources limit\u00e9es. Par cons\u00e9quent, un outil de diagnostic mol\u00e9culaire rapide, pr\u00e9cis et sensible est garanti pour la d\u00e9tection simultan\u00e9e de plusieurs virus au point d\u2019intervention.<\/p>\n<p>Pour combler cette lacune, une \u00e9quipe de scientifiques de la R\u00e9publique de Cor\u00e9e, dirig\u00e9e par le professeur Eunjung Kim de l&rsquo;Universit\u00e9 nationale d&rsquo;Incheon (INU), a r\u00e9cemment d\u00e9velopp\u00e9 un nouveau syst\u00e8me de d\u00e9tection TwinDemic (TDD), con\u00e7u pour la d\u00e9tection simultan\u00e9e du SRAS-CoV et de la grippe A. virus (IAV). Leurs conclusions ont \u00e9t\u00e9 publi\u00e9es dans la revue <i>Capteurs et actionneurs B\u00a0: Chimique<\/i>.<\/p>\n<p>\u00ab\u00a0Le TDD comprend une puce microfluidique transparente en poly (m\u00e9thacrylate de m\u00e9thyle) avec des capteurs de d\u00e9tection de g\u00e8nes \u00e0 base d&rsquo;hydrogel et sans enzyme, ainsi qu&rsquo;un lecteur de fluorescence portable\u00a0\u00bb, explique le professeur Kim en d\u00e9crivant le syst\u00e8me de d\u00e9tection.<\/p>\n<p>Les chambres d&rsquo;hydrogel sont \u00e9quip\u00e9es de sondes personnalis\u00e9es pour d\u00e9tecter les deux agents pathog\u00e8nes viraux cibles\u00a0: le SRAS-CoV-2 (CoV) et l&rsquo;IAV. La r\u00e9action entre l\u2019ADN viral cible et le syst\u00e8me de sonde sp\u00e9cifique amplifie le signal de fluorescence.<\/p>\n<p>Notamment, le syst\u00e8me TDD est facile \u00e0 utiliser, rentable et poss\u00e8de une limite de d\u00e9tection de 0,46 picomolaire (pM) pour le CoV et de 0,39 pM pour l\u2019IAV. Pour confirmer TDD&rsquo;Afin d&rsquo;am\u00e9liorer l&rsquo;efficacit\u00e9 du diagnostic, 15 pr\u00e9l\u00e8vements nasopharyng\u00e9s provenant chacun d&rsquo;individus en bonne sant\u00e9, de patients atteints de COVID-19 et de personnes atteintes de la grippe A ont \u00e9t\u00e9 test\u00e9s.<\/p>\n<p>Pour le diagnostic du COVID-19, le syst\u00e8me TDD a correctement pr\u00e9dit les \u00e9chantillons positifs dans 93,3 % des cas et les \u00e9chantillons n\u00e9gatifs dans 96,7 % des cas.
