La cause est li\u00e9e \u00e0 l&rsquo;expansion d&rsquo;unit\u00e9s d&rsquo;ADN r\u00e9p\u00e9titives, dans lesquelles des cha\u00eenes sp\u00e9cifiques de code g\u00e9n\u00e9tique &#8211; dans ce cas, une s\u00e9rie de nucl\u00e9otides cytosine-ad\u00e9nine-guanine, ou CAG, sur un brin de l&rsquo;ADN et de cytosine-thymine-guanine, ou CTG, sur le volet compl\u00e9mentaire, commence \u00e0 se r\u00e9p\u00e9ter encore et encore, s&rsquo;\u00e9tendant jusqu&rsquo;\u00e0 40 \u00e0 120 copies.<\/p>\n<p>Les copies \u00e9tendues cr\u00e9ent des distorsions dans l\u2019ADN, le rendant plus vuln\u00e9rable aux cassures et aux erreurs de r\u00e9plication. Lorsque les cellules tentent de r\u00e9parer les cassures, des expansions r\u00e9p\u00e9t\u00e9es se produisent, ce qui entra\u00eene la perte des fonctions prot\u00e9iques essentielles, ainsi que la d\u00e9ficience et la mort des cellules nerveuses.<\/p>\n<p>Les recherches du laboratoire de Catherine Freudenreich, professeur et directrice du d\u00e9partement de biologie, ont r\u00e9v\u00e9l\u00e9 un m\u00e9canisme mol\u00e9culaire possible expliquant comment les r\u00e9p\u00e9titions d&rsquo;ADN sont bris\u00e9es puis d\u00e9velopp\u00e9es dans le g\u00e8ne de la maladie de Huntington. Freudenreich et son \u00e9quipe de recherche rapportent les r\u00e9sultats dans une nouvelle \u00e9tude publi\u00e9e dans le <i>Actes de l&rsquo;Acad\u00e9mie nationale des sciences<\/i>.<\/p>\n<p>Selon eux, la cause pourrait \u00eatre des prot\u00e9ines naturelles du syst\u00e8me immunitaire qui ciblent normalement les virus en endommageant leur ADN. Les prot\u00e9ines sont appel\u00e9es prot\u00e9ines catalytiques d\u2019\u00e9dition d\u2019ARNm de l\u2019apolipoprot\u00e9ine B, ou APOBEC.<\/p>\n<p>Les APOBEC font partie d&rsquo;une \u00e9quipe de premi\u00e8re r\u00e9ponse compos\u00e9e de prot\u00e9ines utilis\u00e9es pour combattre l&rsquo;infection virale jusqu&rsquo;\u00e0 ce que la partie adaptative du syst\u00e8me immunitaire puisse g\u00e9n\u00e9rer des anticorps et des cellules T tueuses. Ils fonctionnent en coupant un groupe amine (un azote reli\u00e9 \u00e0 trois hydrog\u00e8nes) des nucl\u00e9otides de cytosine dans l&rsquo;ADN viral simple brin, mut\u00e9 le code g\u00e9n\u00e9tique et rendant finalement les g\u00e8nes viraux inutiles.<\/p>\n<p>Mais dans le cas des personnes atteintes de la maladie de Huntington, certaines de ces prot\u00e9ines auxiliaires peuvent attaquer et d\u00e9stabiliser non pas les virus mais l&rsquo;ADN du patient.<\/p>\n<p>Cela se produit dans des parties de l\u2019ADN humain susceptibles de se transformer en boucles simple brin, comme certains segments d\u2019ADN du g\u00e8ne huntingtin, qui code pour une prot\u00e9ine importante pour de nombreuses fonctions neurologiques. Lorsqu\u2019un APOBEC d\u00e9samine des parties de ce g\u00e8ne contenant la s\u00e9quence CAG\/CTG, il augmente le taux d\u2019erreur des enzymes de r\u00e9paration de l\u2019ADN, les amenant \u00e0 ins\u00e9rer plusieurs CAG\/CTG l\u00e0 o\u00f9 il ne devrait y en avoir qu\u2019un. Cela se produit \u00e0 plusieurs reprises jusqu&rsquo;\u00e0 ce que les r\u00e9p\u00e9titions CAG\/CTG se d\u00e9veloppent suffisamment pour interf\u00e9rer avec la fonction du g\u00e8ne huntingtin.<\/p>\n<p>Il existe 11 types d&rsquo;APOBEC naturels chez l&rsquo;homme, c&rsquo;est pourquoi l&rsquo;\u00e9quipe de recherche de Freudenreich a ins\u00e9r\u00e9 des types d&rsquo;APOBEC uniques dans la levure pour isoler et \u00e9tudier les effets de chacun. APOBEC3A s&rsquo;est av\u00e9r\u00e9 avoir l&rsquo;effet le plus prononc\u00e9, provoquant une expansion des r\u00e9p\u00e9titions d&rsquo;ADN dans un tractus CAG\/CTG, avec un r\u00f4le moindre pour APOBEC3B.<\/p>\n<p>Son collaborateur Steve Roberts, professeur agr\u00e9g\u00e9 \u00e0 l&rsquo;Universit\u00e9 du Vermont, a examin\u00e9 les donn\u00e9es accessibles au public sur l&rsquo;expression des prot\u00e9ines \u00e0 partir d&rsquo;\u00e9chantillons de tissus c\u00e9r\u00e9braux provenant de patients d\u00e9c\u00e9d\u00e9s atteints de la maladie de Huntington et a d\u00e9couvert une pr\u00e9sence inhabituellement \u00e9lev\u00e9e de l&rsquo;enzyme APOBEC3A, confirmant un lien de causalit\u00e9.<\/p>\n<p>\u00ab\u00a0Le niveau est encore plus \u00e9lev\u00e9 que dans les cellules canc\u00e9reuses du sein, o\u00f9 l&rsquo;on sait que les APOBEC jouent un r\u00f4le\u00a0\u00bb, explique Freudenreich. \u00ab\u00a0Cela signifie que les APOBEC sont au bon endroit pour causer des probl\u00e8mes, ce qui en fait des candidats vraiment intrigants pour le d\u00e9clenchement de la maladie.\u00a0\u00bb<\/p>\n<p>Les chercheurs ont trouv\u00e9 des preuves suppl\u00e9mentaires concernant d\u2019autres prot\u00e9ines qui pourraient aider les APOBEC \u00e0 induire les expansions r\u00e9p\u00e9t\u00e9es de l\u2019ADN.<\/p>\n<p>Freudenreich et ses collaborateurs continueront d&rsquo;explorer le r\u00f4le de cette voie dans la maladie de Huntington, ainsi que la question de savoir si les inhibiteurs d&rsquo;APOBEC pourraient freiner leur activit\u00e9.
