Credit: Damiel Marechal\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>Recherche publi\u00e9e dans la revue <i>Glia<\/i> a identifi\u00e9 des liens cruciaux entre les choix alimentaires et la progression de la scl\u00e9rose en plaques (SEP).<\/p>\n<p>L&rsquo;\u00e9tude, dirig\u00e9e par Patrizia Casaccia, directrice fondatrice du Advanced Science Research Center de l&rsquo;Initiative en neurosciences du CUNY Graduate Center (CUNY ASRC) et professeur Einstein de biologie et de biochimie au CUNY Graduate Center, a explor\u00e9 comment les enzymes appel\u00e9es c\u00e9ramide synthase 5 et 6 sont responsable de l&rsquo;effet toxique d&rsquo;un r\u00e9gime riche en huile de palme sur les neurones du syst\u00e8me nerveux central, ce qui entra\u00eene une augmentation ult\u00e9rieure de la gravit\u00e9 des sympt\u00f4mes de la SEP.<\/p>\n<p>La SEP est une maladie auto-immune inflammatoire caract\u00e9ris\u00e9e par des dommages importants \u00e0 la gaine isolante de my\u00e9line qui prot\u00e8ge les nerfs dans tout le corps.<\/p>\n<p>Les traitements actuels se concentrent sur le contr\u00f4le de la r\u00e9ponse du syst\u00e8me immunitaire, mais les m\u00e9canismes pr\u00e9cis contribuant \u00e0 la neurod\u00e9g\u00e9n\u00e9rescence dans la SEP restent mal compris. Des travaux ant\u00e9rieurs du laboratoire Casaccia et d&rsquo;autres avaient fait \u00e9tat de l&rsquo;effet toxique d&rsquo;un r\u00e9gime riche en graisses sur la gravit\u00e9 des sympt\u00f4mes de la SEP. Dans leur \u00e9tude, les chercheurs ont explor\u00e9 les m\u00e9canismes potentiels par lesquels une alimentation riche en huile de palme pourrait nuire \u00e0 la sant\u00e9 neuronale.<\/p>\n<h2>Neuroprotection contre la toxicit\u00e9 induite par l&rsquo;huile de palme<\/h2>\n<p>En utilisant le mod\u00e8le exp\u00e9rimental de d\u00e9my\u00e9linisation inflammatoire de l&rsquo;enc\u00e9phalomy\u00e9lite auto-immune (EAE), l&rsquo;\u00e9quipe de recherche a d\u00e9couvert que les r\u00e9gimes alimentaires riches en huile de palme entra\u00eenaient une \u00e9volution plus grave de la maladie chez la souris.<\/p>\n<p>\u00ab\u00a0Nous avons pens\u00e9 qu&rsquo;\u00e0 l&rsquo;int\u00e9rieur des cellules neuronales, l&rsquo;huile de palme est transform\u00e9e en une substance toxique appel\u00e9e c\u00e9ramide C16 par des enzymes sp\u00e9cifiques appel\u00e9es CerS5 et CerS6\u00a0\u00bb, a d\u00e9clar\u00e9 le chercheur principal Casaccia.<\/p>\n<p>\u00ab\u00a0Ce c\u00e9ramide est responsable des dommages mitochondriaux, qui privent les neurones de l&rsquo;\u00e9nergie dont ils ont besoin pour contrecarrer l&rsquo;inflammation dans le cerveau. Nous avons donc demand\u00e9 si l&rsquo;inactivation de ces enzymes conf\u00e9rerait une neuroprotection.\u00a0\u00bb<\/p>\n<p>Les chercheurs ont d\u00e9couvert qu\u2019en supprimant g\u00e9n\u00e9tiquement les enzymes CerS6 et CerS5 dans les neurones, ils pouvaient pr\u00e9venir la neurod\u00e9g\u00e9n\u00e9rescence dans le mod\u00e8le exp\u00e9rimental de SEP.<\/p>\n<p>\u00ab\u00a0Cela \u00e9tait vrai m\u00eame lorsque les souris \u00e9taient nourries avec un r\u00e9gime riche en acide palmitique\u00a0\u00bb, a d\u00e9clar\u00e9 le co-premier auteur de l&rsquo;article, Damien Mar\u00e9chal, chercheur associ\u00e9 au laboratoire Casaccia. \u00ab\u00a0Ces nouvelles informations mettent en \u00e9vidence une voie m\u00e9tabolique sp\u00e9cifique par laquelle les graisses alimentaires peuvent aggraver les sympt\u00f4mes de la SEP.\u00a0\u00bb<\/p>\n<h2>Importance pour les patients atteints de SEP et les cliniciens<\/h2>\n<p>Les conclusions de l&rsquo;article ont des implications significatives pour les personnes atteintes de SEP ainsi que pour les cliniciens traitant des patients et les neuroscientifiques qui \u00e9tudient la maladie. Ces travaux confirment que les choix de mode de vie, tels que l\u2019alimentation, peuvent avoir un impact profond sur l\u2019\u00e9volution de la maladie.<\/p>\n<p>Les r\u00e9sultats de l&rsquo;\u00e9tude s&rsquo;appuient sur des concepts ant\u00e9rieurs concernant des choix alimentaires judicieux pour g\u00e9rer les sympt\u00f4mes de la SEP.
