Gujarati\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>Une nouvelle recherche men\u00e9e par des sp\u00e9cialistes en n\u00e9phrologie de Stony Brook Medicine, ax\u00e9e sur le ciblage de la signalisation cellulaire cl\u00e9 entre deux types de cellules r\u00e9nales et sur l&rsquo;induction d&rsquo;un certain g\u00e8ne au sein de ces processus, pourrait aider \u00e0 pr\u00e9venir ou \u00e0 r\u00e9duire la progression de la maladie r\u00e9nale diab\u00e9tique (DKD), la principale cause de maladie r\u00e9nale chronique dans le monde.<\/p>\n<p>Selon la National Kidney Foundation, environ 1 adulte sur 7 aux \u00c9tats-Unis souffre d&rsquo;une maladie r\u00e9nale, soit plus de 35 millions de personnes. Jusqu&rsquo;\u00e0 un tiers de tous les Am\u00e9ricains diab\u00e9tiques souffrent d&rsquo;une maladie r\u00e9nale.<\/p>\n<p>Les chercheurs en n\u00e9phrologie doivent encore cr\u00e9er une approche th\u00e9rapeutique pour inverser ou pr\u00e9venir la progression de la DKD. Sandeep K. Mallipattu, MD, et Nehaben A. Gujarati, Ph.D. (auteur principal), de la division de n\u00e9phrologie et d&rsquo;hypertension du d\u00e9partement de m\u00e9decine de la Renaissance School of Medicine de l&rsquo;universit\u00e9 de Stony Brook, esp\u00e8rent que leur mod\u00e8le murin constitue une premi\u00e8re \u00e9tape vers une nouvelle strat\u00e9gie th\u00e9rapeutique plus efficace contre la DKD. Leurs conclusions sont publi\u00e9es dans <i>Communications naturelles<\/i>.<\/p>\n<p>Ils ont utilis\u00e9 une approche multi-omique (g\u00e9nome, prot\u00e9ome, etc.) pour d\u00e9montrer que l&rsquo;induction d&rsquo;un facteur de transcription KLF6 humain et le ciblage de la signalisation cellulaire entre les podocytes et les cellules du tubule proximal att\u00e9nuent la perte de podocytes, le dysfonctionnement du tubule proximal et l&rsquo;\u00e9ventuelle fibrose interstitielle qui survient dans stades ult\u00e9rieurs de la maladie.<\/p>\n<p>Les podocytes jouent un r\u00f4le central dans la r\u00e9gulation de la fonction glom\u00e9rulaire, le processus de filtrage qui \u00e9limine les d\u00e9chets et l&rsquo;exc\u00e8s d&rsquo;eau du sang et forme l&rsquo;urine. Les cellules des tubules proximaux, le type cellulaire pr\u00e9dominant du rein, remplissent de nombreuses fonctions telles que la r\u00e9absorption de l&rsquo;eau, du glucose et de certaines prot\u00e9ines du filtrat glom\u00e9rulaire et maintiennent \u00e9galement l&rsquo;\u00e9quilibre \u00e9lectrolytique et l&rsquo;hom\u00e9ostasie des fluides. Ces deux types de cellules r\u00e9nales sont essentiels pour pr\u00e9venir la progression de la DKD.<\/p>\n<p>Mallipattu et Gujarati ont d\u00e9montr\u00e9 dans ce mod\u00e8le DKD que KLF6 d\u00e9clenche la s\u00e9cr\u00e9tion d&rsquo;apolipoprot\u00e9ine J (ApoJ) par les podocytes, pour amorcer les cellules du tubule proximal en activant la prot\u00e9ine kinase 1D d\u00e9pendante du calcium\/calmoduline (CaMK1D), emp\u00eachant ainsi les l\u00e9sions mitochondriales et arr\u00eatant les processus DKD. .<\/p>\n<p>\u00ab\u00a0Cette communication de cellule \u00e0 cellule via ce m\u00e9canisme de signalisation dans le rein pourrait servir de m\u00e9canisme de protection dans les premiers stades de la DKD\u00a0\u00bb, explique Mallipattu, professeur de m\u00e9decine DCI-Leibowitz et chef de la division de n\u00e9phrologie et d&rsquo;hypertension.<\/p>\n<p>Cette \u00e9tude impliquait une combinaison de mod\u00e8les murins, de cellules humaines et de tissus provenant de biopsies r\u00e9nales humaines provenant de patients atteints de DKD \u00e0 diff\u00e9rents stades.<\/p>\n<p>\u00ab\u00a0En combinaison, les r\u00e9sultats de ces \u00e9tudes mettent en \u00e9vidence que le ciblage de la signalisation des tubules podocytes-proximaux en am\u00e9liorant l&rsquo;apolipoprot\u00e9ine J-CaMK1D pourrait s&rsquo;av\u00e9rer \u00eatre une strat\u00e9gie th\u00e9rapeutique pour ralentir, voire pr\u00e9venir la DKD\u00a0\u00bb, ajoute Mallipattu.<\/p>\n<p>Gujarati et Mallipattu continueront \u00e0 tester cette approche du traitement du DKD.
