{"id":11117,"date":"2025-10-29T03:07:06","date_gmt":"2025-10-29T02:07:06","guid":{"rendered":"https:\/\/rvh-synergie.org\/actualites\/une-etude-animale-relie-les-niveaux-regionaux-de-pfas-aux-impacts-sur-la-reproduction-masculine\/"},"modified":"2025-10-29T03:07:08","modified_gmt":"2025-10-29T02:07:08","slug":"une-etude-animale-relie-les-niveaux-regionaux-de-pfas-aux-impacts-sur-la-reproduction-masculine","status":"publish","type":"post","link":"https:\/\/rvh-synergie.org\/actualites\/une-etude-animale-relie-les-niveaux-regionaux-de-pfas-aux-impacts-sur-la-reproduction-masculine\/","title":{"rendered":"Une \u00e9tude animale relie les niveaux r\u00e9gionaux de PFAS aux impacts sur la reproduction masculine"},"content":{"rendered":"<div>\n<div class=\"article-gallery lightGallery\">\n<div data-thumb=\"https:\/\/scx1.b-cdn.net\/csz\/news\/tmb\/2025\/animal-study-links-reg.jpg\" data-src=\"https:\/\/scx2.b-cdn.net\/gfx\/news\/hires\/2025\/animal-study-links-reg.jpg\" data-sub-html=\"Credit: University of Newcastle\">\n<figure class=\"article-img\">\n<\/figure><\/div>\n<\/div>\n<p>De nouvelles recherches de l&rsquo;Universit\u00e9 de Newcastle ont r\u00e9v\u00e9l\u00e9 que les niveaux de PFAS correspondant \u00e0 ceux trouv\u00e9s dans la zone de contamination de Williamtown pourraient alt\u00e9rer consid\u00e9rablement la sant\u00e9 reproductive des m\u00e2les chez les animaux, m\u00eame sans endommager visiblement les spermatozo\u00efdes.<\/p>\n<p>Dirig\u00e9e par le professeur Brett Nixon de l&rsquo;universit\u00e9 et le Dr Jacinta Martin, du programme d&rsquo;infertilit\u00e9 et de reproduction de l&rsquo;HMRI, l&rsquo;\u00e9tude pr\u00e9clinique a expos\u00e9 des sujets \u00e0 de l&rsquo;eau contamin\u00e9e par des PFAS \u00e0 des concentrations refl\u00e9tant une exposition environnementale r\u00e9elle dans les zones de Williamtown en Nouvelle-Galles du Sud.<\/p>\n<p>Les PFAS (substances per- et polyfluoroalkyles) sont des produits chimiques synth\u00e9tiques qui persistent dans l&rsquo;environnement et s&rsquo;accumulent dans les organismes vivants. De plus en plus de preuves sugg\u00e8rent qu&rsquo;ils peuvent pr\u00e9senter des risques pour la sant\u00e9 humaine, notamment des impacts sur la fertilit\u00e9 masculine. Les derni\u00e8res d\u00e9couvertes chez les animaux soul\u00e8vent des inqui\u00e9tudes quant aux effets potentiels \u00e0 long terme et g\u00e9n\u00e9rationnels de l\u2019exposition aux PFAS.<\/p>\n<p>Publi\u00e9 aujourd&rsquo;hui dans <i>Biologie des communications<\/i>les r\u00e9sultats sont bas\u00e9s sur des \u00e9tudes pr\u00e9cliniques sur des souris et r\u00e9v\u00e8lent\u00a0:<\/p>\n<ul>\n<li>Le nombre de spermatozo\u00efdes en d\u00e9veloppement a chut\u00e9\u00a0: la production quotidienne de spermatozo\u00efdes a diminu\u00e9 pendant la p\u00e9riode d&rsquo;exposition au PFAS.<\/li>\n<li>Les niveaux d\u2019hormones masculines ont chut\u00e9 : la testost\u00e9rone et la DHT (dihydrotestost\u00e9rone ; une puissante hormone androg\u00e8ne d\u00e9riv\u00e9e de la testost\u00e9rone), qui sont vitales pour la production de spermatozo\u00efdes, ont \u00e9t\u00e9 r\u00e9duites.<\/li>\n<li>Les spermatozo\u00efdes portaient des changements cach\u00e9s : les mol\u00e9cules qui aident \u00e0 r\u00e9guler l\u2019expression des g\u00e8nes ont \u00e9t\u00e9 alt\u00e9r\u00e9es.<\/li>\n<li>Le d\u00e9veloppement des embryons a \u00e9t\u00e9 perturb\u00e9 : les premiers embryons pr\u00e9sentaient une expression g\u00e9n\u00e9tique anormale.<\/li>\n<li>Les spermatozo\u00efdes fonctionnaient toujours normalement : ils pouvaient se d\u00e9placer, survivre et f\u00e9conder les ovules dans des conditions de laboratoire, malgr\u00e9 les changements mol\u00e9culaires.<\/li>\n<\/ul>\n<p>L\u2019\u00e9tude met en \u00e9vidence un nouveau m\u00e9canisme potentiel de danger\u00a0: les PFAS peuvent ne pas endommager directement les spermatozo\u00efdes, mais plut\u00f4t modifier les signaux mol\u00e9culaires que transportent les spermatozo\u00efdes, signaux cruciaux pour le d\u00e9veloppement sain de l\u2019embryon.<\/p>\n<p>\u00ab\u00a0C&rsquo;est la premi\u00e8re fois que nous montrons qu&rsquo;une exposition aux PFAS \u00e0 des niveaux pertinents pour l&rsquo;environnement, \u00e9quivalents \u00e0 ceux d\u00e9tect\u00e9s \u00e0 Williamtown, peut modifier la composition mol\u00e9culaire des spermatozo\u00efdes, avec des implications potentielles pour perturber le d\u00e9veloppement de l&#8217;embryon\u00a0\u00bb, a d\u00e9clar\u00e9 le professeur Nixon. \u00ab\u00a0Ce qui est frappant, c&rsquo;est que les spermatozo\u00efdes semblaient toujours fonctionner normalement dans des conditions de laboratoire. Mais sous la surface, ils portaient des changements mol\u00e9culaires qui pourraient affecter la g\u00e9n\u00e9ration suivante.<\/p>\n<p>\u00ab\u00a0Nous avons \u00e9galement constat\u00e9 des niveaux plus faibles de testost\u00e9rone et de DHT chez les souris m\u00e2les expos\u00e9es au PFAS, et la production quotidienne de spermatozo\u00efdes a \u00e9galement diminu\u00e9.\u00a0\u00bb<\/p>\n<p>L&rsquo;\u00e9tude, qui contribue \u00e0 un doctorat. Le projet de l&rsquo;\u00e9tudiante Leah Gillespie a examin\u00e9 les cons\u00e9quences de l&rsquo;exposition aux PFAS sur la fertilit\u00e9 masculine et la sant\u00e9 des embryons. \u00ab\u00a0Nous avons d\u00e9couvert que certains changements dans l&rsquo;expression des g\u00e8nes pourraient avoir un impact sur la sant\u00e9 de l&#8217;embryon\u00a0\u00bb, a d\u00e9clar\u00e9 le Dr Martin. \u00ab\u00a0L&rsquo;un des changements pr\u00e9vus que nous avons remarqu\u00e9s \u00e9tait li\u00e9 \u00e0 la taille du corps et \u00e0 la possibilit\u00e9 que la prog\u00e9niture engendr\u00e9e par des animaux expos\u00e9s aux PFAS naisse ou grandisse de mani\u00e8re significativement plus grande que la normale.\u00a0\u00bb<\/p>\n<p>Ces r\u00e9sultats font \u00e9cho \u00e0 des \u00e9tudes humaines montrant une diminution du nombre de spermatozo\u00efdes chez les hommes fortement expos\u00e9s aux PFAS et sugg\u00e8rent que l&rsquo;exposition paternelle aux PFAS \u00e0 elle seule pourrait avoir des cons\u00e9quences sur les enfants, m\u00eame si les enfants eux-m\u00eames ne sont pas directement expos\u00e9s.<\/p>\n<p>\u00ab\u00a0Notre \u00e9tude \u00e9tait bas\u00e9e sur une exposition environnementale r\u00e9elle.
