Le nouveau compos\u00e9 constitue un \u00e9chafaudage chimique prometteur pour d\u00e9velopper de nouveaux m\u00e9dicaments antituberculeux puissants.<\/p>\n<p>M. tuberculosis, l&rsquo;agent pathog\u00e8ne responsable de la tuberculose (TB), est la principale cause de d\u00e9c\u00e8s li\u00e9s aux maladies bact\u00e9riennes dans le monde. Les sch\u00e9mas th\u00e9rapeutiques antibiotiques actuels pour le traitement de la tuberculose sont obsol\u00e8tes, n\u00e9cessitent des traitements plus longs et risquent de d\u00e9velopper une r\u00e9sistance aux m\u00e9dicaments.<\/p>\n<p>Dans la nouvelle \u00e9tude, les chercheurs ont recherch\u00e9 de nouveaux antibiotiques ciblant M. tuberculosis qui pourraient \u00e9galement \u00eatre efficaces contre les souches r\u00e9sistantes aux m\u00e9dicaments. Dans le domaine de la d\u00e9couverte de m\u00e9dicaments, le monde des compos\u00e9s naturels produits par des organismes tels que les plantes, les champignons et les bact\u00e9ries constitue un point de d\u00e9part pr\u00e9cieux pour commencer \u00e0 rechercher de nouveaux antibiotiques.<\/p>\n<p>La sanguinarine, un compos\u00e9 naturel aux propri\u00e9t\u00e9s antimicrobiennes connues, est extraite d&rsquo;une plante herbac\u00e9e \u00e0 fleurs originaire d&rsquo;Am\u00e9rique du Nord. La sanguinarine a \u00e9t\u00e9 utilis\u00e9e en m\u00e9decine traditionnelle et alternative pour les animaux, mais sa toxicit\u00e9 la rend impropre \u00e0 son utilisation comme m\u00e9dicament chez l&rsquo;homme.<\/p>\n<p>Les chercheurs ont repens\u00e9 la sanguinarine en utilisant les principes de la chimie m\u00e9dicinale pour produire un compos\u00e9 antibact\u00e9rien plus puissant avec une toxicit\u00e9 r\u00e9duite. Dans des \u00e9tudes r\u00e9alis\u00e9es sur des \u00e9prouvettes et sur des souris, la version am\u00e9lior\u00e9e de la sanguinarine, appel\u00e9e BPD-9, \u00e9tait capable de tuer les souches de M. tuberculosis r\u00e9sistantes \u00e0 tous les antibiotiques de premi\u00e8re ligne utilis\u00e9s en clinique pour traiter la tuberculose.<\/p>\n<p>De plus, le BPD-9 s\u2019est r\u00e9v\u00e9l\u00e9 efficace contre M. tuberculosis non r\u00e9plicatif (dormant) et intracellulaire, deux aspects cl\u00e9s qui limitent l\u2019efficacit\u00e9 des m\u00e9dicaments antituberculeux actuels. Les chercheurs ont \u00e9galement d\u00e9couvert que le BPD-9 n\u2019\u00e9tait actif que contre les bact\u00e9ries pathog\u00e8nes du m\u00eame genre que M. tuberculosis, ce qui pourrait \u00e9pargner le microbiome et d\u2019autres bact\u00e9ries b\u00e9n\u00e9fiques que la plupart des antibiotiques nuisent.<\/p>\n<p>\u00ab\u00a0Nos r\u00e9sultats montrent une nouvelle entit\u00e9 chimique qui poss\u00e8de des propri\u00e9t\u00e9s uniques dans la lutte contre Mycobacterium tuberculosis, qui pourraient \u00eatre exploit\u00e9es davantage \u00e0 des fins cliniques\u00a0\u00bb, a d\u00e9clar\u00e9 l&rsquo;auteur correspondant de l&rsquo;\u00e9tude, Jim Sun, Ph.D., professeur adjoint au d\u00e9partement de microbiologie et d&rsquo;immunologie de l&rsquo;universit\u00e9.
